The β-galactosidase activity of 15 Kluyveromyces strains isolated from cheese belonging to Kluyveromyces lactis and Kluyveromyces marxianus species was tested for the production of oligosaccharides derived from lactose (GOS) and lactulose (OsLu). All Kluyveromyces crude cell extracts (CEEs) produced GOS, such as 6-galactobiose and 3'-, 4'-, and 6'-galactosyl-lactose. At 4 h of reaction, the main trisaccharide formed was 6'-galactosyl-lactose (20 g/100 g of total carbohydrates). The formation of OsLu was also observed by all CEEs tested, with 6-galactobiose, 6'-galactosyl-lactulose, and 1-galactosyl-lactulose being found in all of the reaction mixtures. The synthesis of trisaccharides predominated over other oligosaccharides. K. marxianus strain O3 produced the highest yields of GOS and OsLu after 4 h of reaction, reaching 42 g/100 g of total carbohydrates (corresponding to 80% lactose hydrolysis) and 45 g/100 g of total carbohydrates (corresponding to 87% lactulose hydrolysis), respectively. Therefore, the present study contributes to a better insight into dairy Kluyveromyces β-galactosidases and shows the feasibility of these enzymes to transglycosylate lactose and lactulose, producing high yields of prebiotic oligosaccharides.  相似文献

Since swine wastewater is used by farmers for soil fertilization, evaluation of toxic compounds or micro-contaminants of separate streams is required. This paper uses the toxicity identification evaluation (TIE) procedure for the physicochemical and ecotoxicological characterization of swine wastewater. To distinguish the most important toxic compounds, a physicochemical characterization and phase I-TIE procedure were performed. The acute toxic effect of swine wastewater and treated fractions (phase II-TIE) were evaluated using Daphnia magna determining 48-h LC50. Results show a high level of conductivity (23.5 μS cm−1), which is explained as due to the concentration of ions, such as ammonium (NH4+–N 1.6 g L−1), sulfate (SO42− 397.3 mg L−1), and chlorine (Cl 1,230.0 mg L−1). The acute toxicity of the swine wastewater was evaluated on D. magna (48-h LC50 = 3.4%). Results of the different water treatments indicate that anionic exchange treatments could reduce 22.5% of swine wastewater’s acute toxicity by reducing chlorine (to around 51%) and conductivity (8.5%). On the other hand, cationic exchange treatment increased acute toxicity on D. magna (% RT = −624.4%), by reducing NH4+–N (around 100%) and total nitrogen (95.5%). This finding suggests that part of the toxicity comes from anionic compounds, such as chlorine.  相似文献

We here isolate fulvic acids from vermicompost to prepare and characterize novel fulvic acid-coated magnetite nanoparticles. UV-A irradiation of suspensions of the nanoparticles under different experimental conditions led to photo-reduction of Cr(VI). In anoxic conditions in the presence of formic acid, after 60 min of irradiation ca. 100% of Cr(VI) was reduced. Under these conditions, the carbon dioxide radical anions, CO2 .- , mediated the photo-reduction of Cr(VI). However, the high reduction potential of Cr(VI) and the variety of reactive species generated upon UV-A irradiation make this nanomaterial also suitable for Cr(VI) photo-reduction also under aerobic conditions in the presence of formic acid or under anoxic conditions without the addition of formic acid. To address the drivers of biodiversity loss, conservation efforts must attend to public attitudes toward endangered species. Using sea otters as a species at risk, we examined how messaging shapes people’s attitudes toward this species. Participants viewed a message about sea otters that was either positive (sea otters as keystone species), negative (resource conflict with local fishermen), or neutral (biological facts). The impact of the message on people’s attitudes was measured using Kellert’s typology of basic attitudes toward wildlife. We found that the negative message promoted people’s interests in the practical value of sea otters (utilitarian-consumption attitudes), their habitats (utilitarian-habitat attitudes), and control over sea otters (dominionistic attitudes), even though the negative message was perceived as less convincing and believable than positive or neutral messages. The positive message decreased utilitarian-consumption attitudes, and the neutral message decreased utilitarian-habitat attitudes. Our findings suggested that messaging can influence public attitudes toward wildlife.  相似文献

Bluetongue virus (BTV) is a double-stranded RNA virus that induces apoptosis both in mammalian cell cultures and in target tissues. Based on information that members of the mitogen-activated protein kinase family (MAPKs) are mediators of apoptosis, we have examined in detail the MAPK-dependent apoptosis in BTV infection. Previously, we have shown that apoptosis in BTV infection requires the participation of mitochondrial apoptotic pathways. In addition, we demonstrated that NF-κB is activated and that its inhibition substantially reduces cellular apoptosis. For the first time, here we demonstrated the activation of MAPKs after BTV infection. Moreover, by pre-treatment with MAPK inhibitors, c-Jun N-terminal kinases (JNKs) and p38 MAPK, but not extracellular signal-related kinase (ERK), significantly decreased the induction of apoptosis. JNK and p38 activation regulated the cytochrome c released from mitochondria and caspase 3 activation. These results strengthen the understanding of BTV infection and contribute to our previous data confirming that BTV infection induces robust apoptosis in mammalian cells and is likely to play a primary role in BTV pathophysiology.  相似文献

The presence of parasites was investigated by the examination of 1944 dog faecal samples collected from urban (n = 646) and rural (n = 1298) areas of the province of Neuquén, Patagonia, Argentina. Parasitic agents (PA) were found in 37.86% of samples. A total of 15 different PA were detected, including Toxocara canis (16.35%), Taenia spp./Echinococcus spp. (12.65%), Trichurisvulpis (6.06%), Giardia spp. (1.29%), Toxascaris leonina (0.56%), Ancylostomacaninum (0.41%), Dipylidium caninum (0.31%), Diphyllobothrium spp. (0.10%), among others. Several of these PA are recognized as zoonotic agents. Therefore, the results of this investigation revealed that local population is exposed to a broad spectrum of zoonotic parasites by means of environmental contamination with dog faeces. Prevalence of PA was slightly higher in rural (40.06%) than in urban (33.44%) locations. Distribution of groups of PA (cestodes, nematodes, and protozoa) showed statistical differences between both habitats. Prevalence of cestodes (18.18%) and protozoa (11.86%) was significantly higher in the rural environment than in urban areas and nematodes (29.10%) were more frequent in urban locations. Infection of dogs with Linguatula serrata and Cryptosporidium sp. was demonstrated for the first time in Neuquén. Rural dogs of the study area are under hydatic disease control program, which includes treatment with praziquantel every 6 weeks; thus, the finding of high level of cestode infection in these areas is of great relevance. The epidemiology of zoonotic parasitic infections in urban and rural dogs showed different patterns and, in consequence, different control measurements should be applied in each location.  相似文献

Finkel-Biskis-Reilly murine sarcoma virus (FBR-MuSV) ubiquitously expressed (FAU) gene is down-regulated in human prostate, breast and ovarian cancers. Moreover, its dysregulation is associated with poor prognosis in breast cancer. Sponges (Porifera) are animals without tissues which branched off first from the common ancestor of all metazoans. A large majority of genes implicated in human cancers have their homologues in the sponge genome. Our study suggests that FAU gene from the sponge Suberites domuncula reflects characteristics of the FAU gene from the metazoan ancestor, which have changed only slightly during the course of animal evolution. We found pro-apoptotic activity of sponge FAU protein. The same as its human homologue, sponge FAU increases apoptosis in human HEK293T cells. This indicates that the biological functions of FAU, usually associated with “higher” metazoans, particularly in cancer etiology, possess a biochemical background established early in metazoan evolution. The ancestor of all animals possibly possessed FAU protein with the structure and function similar to evolutionarily more recent versions of the protein, even before the appearance of true tissues and the origin of tumors and metastasis. It provides an opportunity to use pre-bilaterian animals as a simpler model for studying complex interactions in human cancerogenesis.  相似文献
